Orthogonal Measures of Lentiviral Particle Attributes Using DLS, NTA, and Micro Flow Cytometry. Lentiviral vectors are used in multiple cell and gene therapy programs, both ex vivo and in vivo. During production, enveloped lentiviral particles are produced along with other membrane-bound particles that may or may not have similar protein content and other properties. Throughout the production, purification, formulation, and storage of these products, monitoring the quantity, size distribution, charge, and protein content can be helpful. There are many different instruments and methodologies that facilitate these analyses, such as dynamic light scattering, nanoparticle tracking analysis, flow virometry, and protein analysis by ELISA and Western Blot. This talk will cover the utility of using these and other analytical methods to assist product developers who seek to implement robust processes that reproducibly produce high-quality products.

Jim leads the analytical development group at Interior. Trained as a virologist, Jim has over 25 years of experience in developing viral products for the prevention and treatment of disease. Jim comes to Interius from Altimmune, where he was Director of Scientific Affairs, leading efforts to identify and evaluate strategic opportunities. Jim also led the cell and gene therapy standards development efforts at US Pharmacopeia. In previous roles at Advanced BioScience Laboratories and the Foundation Fighting Blindness, he led translational science activities for the development of vaccines and biologics to prevent and treat infectious and retinal diseases. Jim has also held positions responsible for performing viral clearance and adventitious agent testing at Viromed Biosafety as well as AAV vector development and characterization at Genovo/Targeted Genetics. Dr Richardson earned his Ph.D. in Biomedical Sciences at the Mount Sinai/Icahn School of Medicine.
Kevin Lance, PhD, Director of Viral Vector Marketing at Unchained Labs
Kevin Lance, PhD, is the Director of Viral Vector Marketing at Unchained Labs. His expertise covers technologies like DLS, UV/Vis, DSF, and others. He has experience in the biophysical characterization of proteins, nucleic acids, viral vectors, and LNPs. Before Unchained Labs, his research experience covered the fields of sustained antibody delivery and nanotechnology. He earned his Ph.D. in bioengineering from the joint UC Berkeley-UCSF Bioengineering Graduate Program.
