The promising success stories and advancements of viral vector-based gene therapy are emblematic of the future of Gene therapy. With the promising future, we still see multiple setbacks and technological challenges towards manufacturing and the higher cost of drug products, which makes it clear that technological challenges must still be overcome, working together with the gene therapy industry.

The viral vector production and purification processes are widely divided through the use of different technologies and tools available across industries. Therefore, a platform approach will be beneficial for harmonizing production processes for multiple viral vectors, which will help optimize manufacturing efforts, thus reducing the cost of goods for manufacturing.

Adeno-associated virus (AAV) and Lentivirus (LV) are two major viral vectors being used for in-vivo and ex-vivo gene therapies respectively, which have unique manufacturing challenges of their own. Based on the multiple serotypes of AAV or multiple pseudo-typing strategies for Lentivirus, the process development is challenging. In this webinar, we will provide an overview of technologies available for the production and downstream purification of AAV & LV vectors and how utilizing the platform approach can help in optimization and reducing the cost of manufacturing.

Ashish Saksule is a cell and gene therapy process development expert focusing on the bioprocessing of viral vectors. He enjoys working on both upstream and downstream processing for viral vectors and has end-to-end production experience for lentiviruses, AAV, live viruses, and proteins.

Ashish has more than 8 years of experience, spanning academic, clinical stage vaccine research, process development for cell and gene therapy, pre-clinical development, and manufacturing. He also holds two Master’s degrees: a Master’s ALM in Biotechnology from Harvard University and an M.S. in Chemical Engineering from Michigan Technological University.

Currently, he is working at Vertex Pharmaceuticals Cell and Genetic Therapy Department, leading the Vector Core facility. He previously worked at Takeda in the Rare Disease Gene Therapy program as a Vector Production Lead and at MilliporeSigma on virus and gene therapy process development for AAV & LV. His other previous experience includes working with Lentigen-Miltenyi Biotec for process development and preclinical production of Lentivirus and at Novavax.

Ashish is interested in a vector platform process approach, production optimization, and reducing the cost of the drug for in vivo and ex vivo gene therapy treatments.

Dr. Alejandro Becerra is a Principal Applications Scientist and Global Purification Technical Lead at Thermo Fisher Scientific. Alejandro is a chemical engineer by training and holds a Ph.D. from Cornell University.  Dr. Becerra has over 15 years of experience in downstream processing and customer support, having spent 11 years at Millipore in various bioprocess engineering roles before joining Thermo Fisher in 2018. Alejandro is a subject matter expert in preparative chromatography with expertise in the development, optimization, and scale-up of purification processes for antibodies, recombinant proteins, viral vectors, and nucleic acids.
